She was born in Portland, Maine, the daughter of William K and Janice Kendrick. Her descent is Scottish, English, and Irish. Her older brother is actor Michael Cooke Kendrick, who appeared in the 2000 movie Looking for an Echo. She attended Deering High School in her hometown.

Anna began her performing career at the early age of 10 when she caught a bus from her hometown to New York City where she could try out for roles. Kendrick's first role was as Dinah in the Broadway production High Society in August 1998 when she was 12 years old, a role for which she won the Theatre World Award. Also, she was nominated for a Tony Award and a Drama Desk Award. The Tony Award nomination made her the third-youngest Tony Award nominee ever. Anna went on to appear in a number of other stage productions, including A Little Night Music, playing Fredrika Armfeldt in the New York City Opera production. A little later, she made her film debut in the 2003 comedy musical Camp. Due to her portrayal of Fritzi Wagner in this film, she received a nomination for the Independent Spirit Awards Best Debut Performance.

In 2007, Anna appeared in her next film, Rocket Science, in which she portrayed Ginny Ryerson, a fast-talking high school debater. Although she said she found the role intimidating and challenging after watching an actual national collegiate championship debate, her performance was praised by critics and she was nominated for the Independent Spirit award for Best Supporting Actress. Later that year, Anna auditioned for the part of Jessica Stanley in Twilight, a 2008 film based on the initial book of the series by Stephenie Meyer. She had intended to do auditions with various actors, but she was too ill and had to leave prematurely. Fortunately for her, she was able to come back for a later session and was given the role of Jessica. Kendrick reprised this role in the first three Twilight sequels.

Anna's theater singing experience gave her the opportunity to land a role in The Marc Pease Experience (2009), where she appears as a high school senior involved in musical theater. She also appeared in the 2009 film Elsewhere, in which she portrays her first lead character, a girl whose best friend goes missing. She also appeared in the movie Up in the Air, based on Walter Kirn's 2001 book of the same name. For her performance in Up in the Air, the National Board of Review named her Best Supporting Actress, and she also garnered nominations from numerous other awards including The Golden Globes, The Screen Actors Guild, and the Academy Awards.

Kendrick played Stacey Pilgrim, the sister of the lead character. In 2010, she appeared in LCD Soundsystem's music video "Pow Pow" as a "shape-shifter" who gathers "the souls of wicked men". In October 2010, she attended the 2010 Top Glamour Awards based in Mexico where she won the award for "Best International Actress".
