Anna was born in the state of Maine, the daughter of William K and Janice Kendrick. She is of English, Irish, and Scottish descent. Her older brother is actor Michael Cooke Kendrick, who appeared in the 2000 production Looking for an Echo. She attended Deering High School in her hometown.

Anna started her career at the age of 10 when she caught a bus from Portland, Maine, to New York City where she could try out for roles. Kendrick's first role was as Dinah in the Broadway musical High Society in August 1998 when she was only 12 years old, a performance for which she was awarded the Theatre World Award. She was also nominated for a Tony Award and a Drama Desk Award. The Tony Award nomination made her the third-youngest Tony Award nominee ever. Kendrick went on to appear in a number of other stage productions, including the musical A Little Night Music, playing Fredrika Armfeldt in the New York City Opera production. Afterwards, she made her motion picture debut in the 2003 comedy musical Camp. Due to her portrayal of Fritzi Wagner in this film, she received a nomination for the Independent Spirit Awards Best Debut Performance.

In 2007, she appeared in her next film, Rocket Science, in which she played Ginny Ryerson, a fast-talking high school debater. Although Kendrick said she found the role intimidating and challenging after watching a real national collegiate championship debate, her performance was given high accolades by critics and she was nominated for the Independent Spirit award for Best Supporting Actress. Later that year, she tried out for the role of Jessica Stanley in the film adaptation of Twilight, a 2008 film based on the first book of the series by Stephenie Meyer. She had intended to do a mix-and-match audition with various actors, but she was too sick and had to leave. Fortunately for her, she was able to come back for a later session and won the role of Jessica. Kendrick reprised this role in the first three Twilight sequels.

Anna's singing experience in theater gave her the opportunity to star in The Marc Pease Experience (2009), where she appears as a high school senior involved in musical theater. She also appeared in the 2009 film Elsewhere, in which she portrays her first lead character, a girl whose best friend goes missing. She also starred in Up in the Air, based on Walter Kirn's 2001 book of the same name. For her performance in Up in the Air, the National Board of Review named her Best Supporting Actress, and she also garnered nominations from numerous other awards including The Golden Globes, The Screen Actors Guild, and the Academy Awards.

Kendrick reprised her role as Jessica Stanley in The Twilight Saga: Eclipse. She was featured in Edgar Wright's rendition of the graphic novel Scott Pilgrim, titled Scott Pilgrim vs. the World. In this film, she played Stacey Pilgrim, the sister of the lead character. In 2010, she appeared in LCD Soundsystem's music video "Pow Pow" as a "shape-shifter" who gathers "the souls of wicked men". In October 2010, she attended the 2010 Top Glamour Awards in Mexico where she won the award for "Best International Actress".

In 2011, she was featured in the dramatic comedy 50/50, and reprised her role as Jessica Stanley in The Twilight Saga: Breaking Dawn. In 2012, she was Rosie in the film version of the baby book What to Expect When You're Expecting. She also voiced the character Courtney in the stop-motion animated film ParaNorman. She starred in Pitch Perfect as Beca, a rebellious college student who learns that she has a voice in her school's all-women a cappella group, and she is also featured on the film's soundtrack. Kendrick also appeared in the police drama End of Watch, portraying Janet. In 2012 she was in the thriller The Company You Keep.

In 2013, she scored a popular song with "Cups (When I'm Gone)" from the movie Pitch Perfect which became a No. 2 hit on Billboard's Adult Pop chart. Anna also made her SNL hosting debut on April 5, 2014 with musical guest Pharrell.
